What is the correct hamartiological doctrine?

According to Wikipedia: hamartiology is the study of sin with a view to articulating a doctrine of it. In other words, What is sin, its nature and origin?

As far as I’m concerned, "sin" means knowingly doing something to harm someone - either myself, or someone else. Whether that is "correct" or not appears to me to be quite subjective, but it’s a definition that works for me.
